C–C activation reactions have attracted significant interest in the past decades due to their efficiency in constructing or editing complex molecular backbones. Current C–C activation transformations mainly include strained rings, pre-aromatized intermediates, forming strong M–CN bonds or β-scission to give stable products. These requirements in substrate patterns largely limit their further applications. Therefore, novel strategies to selective activate C–C bonds in more general substrates are still highly sought after. In this seminar, I will introduce my work on the strategic manipulation of C–C bonds in unstrained ketones and amides. With rationally designed activation reagents and ligands, we achieved the efficient structural editing for general ketones and amides, including annulation, homologation, and ring contraction. These transformations offer new strategies for chemical synthesis and broaden the space for derivatization of bioactive molecules.
